Studies on the Constituents Analysis for Commercial Rehmanniae Radix Preparata


Abstract
In order to obtain the basal data for the quality control in a commercial oriental medicine. the constituents of Rehmanniae Radix Preparata, such as capapol, 5-HMF and the carbohydrates, were analyzed. The results were :
1. The catapol concentrations of eight commercial Rehmanniae Radix Preparatas were all below 1.5§·/g radix.
2. The 5-HMF concentrations of the commercials was 0.518-3.485 §¶/g radix, and the average was 1.502 §¶/g radix. Among eight commercials, one had a similar content up to 5th Rehmanniae Radix Preparata in the concentration of 5-HMF, and two had similar content up to 5th Rehmanniae Radix Preparata in the concentration of 5-HMF, and two had similar contents up to 4th, and five had similar contents up to 2nd.
3. The commercials had all the same composition in the kinds of carbohydrate, and had similar contents in fructose. raffinose and stachyose, and had no rhamnose. These results suggested that the commercials were produced by a way of processing the above twice.
4. The results, obtained from the carbohydrate analysis by TLC and from the determination of catapol, were not sufficient to determine the number of processing time for the commercials. And it was roughly possible with the concentration of 5-HMF to calculate the number, but was not definitive.
Thus, it was required to control the quality of commercial Rehmanniae Radix Preparata to establish the quantitative controls for the carbohydrates and steroids or to examine the effect of previously reported compound.
